Inclusion criteria

Inclusion criteria: Women diagnosed with infertility who undergo in vitro fertilization (IVF) or intracytoplasmic sperm injection (ICSI) treatment and have blastocysts undergoing preimplantation genetic testing for aneuploidy (PGT-A). Women undergoing controlled ovarian stimulation using a GnRH antagonist protocol, consisting of recombinant follicle-stimulating hormone (rFSH) for ovarian stimulation, GnRH antagonist for prevention of premature ovulation, and dual triggering with GnRH agonist and recombinant human chorionic gonadotropin (hCG) for final oocyte maturation

Exclusion criteria

Exclusion criteria: Women with uterine cavity abnormalities that may affect implantation or pregnancy outcomes, including endometrial polyps or submucosal uterine fibroids. Women diagnosed with conditions known to influence gene expression profiles, including endometriosis or polycystic ovary syndrome (PCOS). Women with a body mass index (BMI) greater than or equal to 30. Women whose embryos fail to develop to the blastocyst stage. Embryos with an insufficient number of trophectoderm cells available for gene expression analysis.

Design outcomes

Primary

MeasureTime frame
Differential expression of DNA repair-related genes between euploid and aneuploid blastocysts Relative gene expression levels measured by RNA sequencing (RNA-seq) and validated by RT-qPCR. Expression reported as fold change and delta-delta Ct values. Relative gene expression levels measured by RNA sequencing (RNA-seq) and validated by RT-qPCR. Expression reported as fold change and delta-delta Ct values

Secondary

MeasureTime frame
Implantation rate 14 days after embryo transfer Presence of serum beta-hCG following transfer of a single euploid embryo,Clinical pregnancy rate 6 weeks of gestation Presence of intrauterine gestational sac with fetal heartbeat detected by transvaginal ultrasonography,Miscarriage rate Up to 20 weeks of gestation Pregnancy loss after confirmation of clinical pregnancy
