Inclusion criteria

Inclusion criteria: Being female. At least 60 years of age. Be physically independent. Have a body mass index greater than 24.9 kg / m². Obtain a minimum score of 14 on the mini mental state exam (FOLSTEIN et al., 1975). Absence of musculoskeletal or cardiovascular disorders that make it impossible to perform high-intensity physical exercises. Be at least six months without performing any regular physical activity.

Exclusion criteria

Exclusion criteria: Attendance rate below 80%. Miss any of the evaluation sessions. Begin some type of regular exercise practice in addition to that proposed in this work during the intervention period.

The aging of the immune system will be measured by flow cytometry, through the collection of peripheral blood mononuclear cells (PBMCs) from the volunteers, which will be separated by Fycoll-Hipaque gradient centrifugation (Pharmacia AB, Upsalla, Sweden) before and after the training protocol. PBMCs will be counted and adjusted to a concentration of 2 x 107 cells / mL in RPMI 1640 medium (Gibco, Grand Island, New York, USA). Aliquots of 100 µl of cell suspension will be plated in 96-well plates (1 x 106 cells / well). The cells will be centrifuged and immediately transferred to cold RPMI 1640 (Sigma) medium containing 10% SFB and kept at 4 ºC until use. After this process, they will be incubated with 50 µl of blocking solution for 20 minutes at 4 ºC, composed by RPMI 1640 medium (Sigma) containing 10% inactivated sheep serum (Fc receptor blocking) and 10% fetal bovine serum (Gibco ). For analysis of the different types of T cells, the samples will be incubated with 20 µl of anti-human monoclonal antibodies, conjugated to fluorochromes for 30 min. Antibodies CD4FITC, CD8PE will be used to identify T lymphocytes and antibodies CD57APC, CD28APCcy7 and KLRG1PercP for senescence analysis. After incubation, the samples will be washed twice in RPMI 1640 medium (Sigma), fixed in 2% formaldehyde (Merck) and kept in the refrigerator until reading when they will be washed twice in PBS. The cells will be analyzed in an Attune NxT flow cytometer and the data analysis will be performed using the FlowJo software. A reduction in CD57 and KLRG1 expression and an increase in CD28 expression are expected.;Inflammatory responses will be assessed by immunoassay (ELISA) by centrifuging blood samples for 10 minutes at 1370xg to separate the plasma, which will be stored at - 20 ° C until analysis. The nutritional assessment will be carried out with the participants who will be instructed by an experienced nutritionist to record their food consumption in a food record of three non-consecutive days (two days a week and one day at the weekend) following the evaluation moments shown in the figure 1. To increase the accuracy of the measurements, recommendations will be given on how to register and how to identify portions. Based on the recalls, the consumption of macronutrients (proteins, carbohydrates and lipids) and energy consumption will be quantified through the Avanutri 3.1.4 software (Avanutri Processor Software, Rio de Janeiro, Brazil; Version 3.1.4). Participants were instructed to maintain their eating habits throughout the intervention. It is expected that there will be no changes in the eating pattern.;The global functionality will be measured through the "Gallon-jug shelf-transfer" test in which the volunteer will stand next to a cupboard with several shelves where five gallons filled with water (? 3.99 kg) will be placed on the shelf that will be at the height of the subject's patella and the subject should transfer the gallons one at a time to a shelf that will be positioned at the height of the subject's shoulder. The test is measured by time, starts with the command of the appraiser and is finished as soon as the last gallon is transferred. To minimize interference, the subject will always use the same hand and use the lower limbs to start the movement (SIGNORILE et al., 2007).which will assess the time that the individual spends to sit and get up from a chair five times, understanding that this measure is associated with the power of the lower limbs and the reaction capacity of the elderly woman. The test will be timed from the evaluator's command until the subject sits and gets up from the chair (without using the arms) five times.
