Interventions

Participants will be randomly assigned to consume either 1) an energy-balanced diet (100% of daily recommended energy intake, 1.2 g protein/kg BW/d), 2) a 40% energy-restricted diet (60% of daily reco
The dietary intervention will be 7 days in duration. In addition, all participants will undergo 7 days of single-leg cast immobilization.

Age
18 Years to 64 Years

Inclusion criteria

Inclusion criteria: Male sex Aged 18-35 years Body mass index (BMI) 22-30 kg/m2

Exclusion criteria

Exclusion criteria: History of medical (including but not limited to gastrointestinal, musculoskeletal, metabolic) disorders which might interfere with the execution of the experiment or influence the study outcomes (to be decided by the investigator and responsible physician)(Family) history of deep vein thrombosis, Factor V Leiden, or other known thrombophilia such as protein C, protein S, antithrombin deficiency  Lower limb, back or shoulder injuries which may interfere with the use of crutches Use of any medications known to affect protein metabolism, including but not limited to chronic use of corticosteroids, non-steroidal anti-inflammatory drugsUse of anti-coagulantsStructural (habitual) and continued (during the intervention period) use of supplements known to affect protein metabolism, including but not limited to protein, (branched chain) amino acids, creatine, and omega-3Food allergies or intolerances to the diets that are providedSmoking Any implants that would be a contra-indication for performing an MRI scan

MeasureTime frame
The primary study parameter is daily integrated myofibrillar protein synthesis rate (%/day). 

Secondary

MeasureTime frame
Secondary study parameters include leg skeletal muscle volume and maximal leg muscle strength.
