Eligibility
Inclusion criteria
Inclusion criteria: Mentally competent older adults who are member of Hersenonderzoek.nl and self-reported to have no cognitive deficits or self-report to have AD. This will be assessed based on the questionnaire filled in by the participants during their registration in Hersenonderzoek.nl, and through the hersenonderzoek AD campaign
Exclusion criteria
Exclusion criteria: • Adults will be excluded if they are mentally incompetent, i.e., have severe dementia. • Adults will be excluded if they, have diabetes, renal or hepatic disease. • Adults taking the following medications will be excluded: anti diabetic medications, systemic glucocorticoids, antipsychotics. • Adults who are vegan or allergic to any of the dietary constituents of the meal intervention will be excluded. • Adults who are contraindicated to venipuncture.

Primary
| Measure | Time frame |
|---|---|
| The main study endpoint will be the influence of food intake on the concentrations of plasma protein biomarkers related to AD or other neurological/neurocognitive diseases. Initial biomarkers of interest are Aβ42, Aβ40, NfL, GFAP and different total tau and phosphorylated tau isoform, and in addition novel biomarkers identified with proteomics. | — |
Secondary
| Measure | Time frame |
|---|---|
| The confounding effect of an overall health status reflected by clinical routine blood analysis and medical history taking on the effect of fasting and food consumption on the levels of BBMs will be investigated. This is based on the hypothesis that overall health could impact metabolism and as such might indirectly impact how food intake influences the levels of BBMs. Measurements include fasting blood glucose, serum concentrations of creatinine, uric acid, urea and c-reactive proteins. All will be analysed using the fully automated Cobas C 503 (Roche Diagnostics GmbH, Penzberg, Germany) at the department of Laboratory Medicine, Amsterdam UMC. Parameters age, sex, body composition (weight/height/waist circumference) and blood pressure will be assessed also. We will also collect medical history and any dietary supplement and/or medication use. In addition, biological variation in plasma AD biomarkers can be assessed in the current study. | — |
