Inclusion criteria
Inclusion criteria: children requiring (laryngo-tracheo-)brochoscopy for clinical indications scopy will take place during office hours
Exclusion criteria
Exclusion criteria: children with malignancies requiring bronchoscopychildren who have had a (lung) transplantation in the past.Children younger than 1 year old: the diameter of their airway does not allow introduction of a bronchoscope with sufficient size working channel for the biopsy forceps and the higher risk of anaesthesia in this groupChildren and parents who cannot read the Dutch patient information.

| • Comparative genomics of nasal and bronchial epithelial cells in children. Using gene expression and epigenetic DNA markers of epithelial cells of the upper and lower airways, we will describe which markers of the upper airway (nose) can be used as a valid reflection of the lower airways by validating them against matched single cell RNA seq of airway wall biopsies. We will publish a catalogue of gene expression signatures in children that reflect the bronchial epithelium that can be used by researchers all across the world. • Establishment of an Epithelial Organoid Repository for mechanistic and interventional pediatric respiratory research. • To describe the single cell composition of the airway wall in pediatric respiratory disease, including asthma, severe wheeze, and cystic fibrosis (CF). As airway wall biopsies contain 50 - 70 % epithelial cells, this may serve as an excellent validation of our epithelial organoids and provide more insight into the cell types and states of pediatric respiratory disease. | — |
