Interventions

Subjects will be asked to take two pills of nicotinamide riboside (250 mg/pill), or placebo, twice daily (two with breakfast and two with lunch, a total of 4 pills/day), for 40 days.

Age
18 Years to 64 Years

Inclusion criteria

Inclusion criteria: Caucasian Males and postmenopausal females Aged 45-65 years at start of the study Body mass index (BMI) 27 * 35 kg/m2 Stable dietary habits (no weight loss or gain >5kg in the past 3 months) Sedentary lifestyle (not more than 2 hours of intensive exercise per week)

Exclusion criteria

Exclusion criteria: Type 2 diabetes Active diseases (cardiovascular, diabetes, liver, kidney, cancer or other) Contra-indication for MRI Participation in earlier research or medical examination that included PET/CT scanning Alcohol consumption of >2 servings per day Smoking in the past 6 months

Hepatic and whole body insulin sensitivity.

Secondary

MeasureTime frame
Muscle mitochondrial function. Ectopic lipid accumulation (intrahepatic and intramuscular). Brown adipose tissue activity. Cardiovascular risk parameters: cardiac energy status, 24h average systolic and diastolic blood pressure, plasma cholesterol levels (cholesterol, HDL, LDL, lathosterol), plasma vasodilatation markers (VCAM1, ICAM1, E-selectin, nitrite) and plasma markers of low grade inflammation (TNF*, interleukins). Whole-body energy expenditure. Body composition (fat mass and fat free mass). Acetylcarnitine levels.
