Age
18 Years to 99 Years

Inclusion criteria

Inclusion criteria: - Lean men (n=10; normoglycemic BMI 20-25 kg/m2) - Obese men (n=10 insulin resistant and n=10 insulin sensitive based on HOMA; BMI 30-35 kg/m2) - Aged 45-65 years - Caucasian - Normal blood pressure (systolic blood pressure 100-140 mmHg, diastolic blood pressure 60-90 mmHg) - Weight stable in last 3 months (± 2kg).

Exclusion criteria

Exclusion criteria: - Smokers - People with intensive fitness training, eg. athletes (>= 3 per week >= 1 hour training) - Serious pulmonary, cardiovascular, hepatic or renal disease - History of cardiovascular disease - Bleeding disorders - Current use of medication interfering with study endpoints/hypotheses (e.g. beta-blockers) - Not to be able to understand the study information - Subjects on a special diet or vegetarian - Blood donation 2 months prior to the study and during the study - Participation in other studies

Design outcomes

Primary

MeasureTime frame
The main study parameter is the effect of selective knockdown on glycerol release in the medium (as marker of lipolysis), TAG and DAG lipase activity, and lipid content in human adipocyte and myocyte cell cultures.

Secondary

MeasureTime frame
As secondary endpoints, differences in localization and trafficking of lipolytic proteins will be investigated in adipocytes derived from obese insulin resistant versus insulin sensitive men.
