Inclusion criteria

Inclusion criteria: (1) Inclusion Criteria for the Prediabetes Group: 1) Meet the diagnostic criteria for prediabetes as defined by the 2023 edition of the Chinese Expert Consensus on the Intervention for Prediabetes in Adults. 2) Age: 18 to 70 years. 3) Not currently taking any other glucose-lowering or lipid-lowering medications. 4) Willing to participate in the study, capable of providing signed informed consent, and able to undergo the treatments and all required examinations. (2) Inclusion Criteria for the Type 2 Diabetes Mellitus (T2DM) Group: 1) Meet the diagnostic criteria for Type 2 Diabetes Mellitus as outlined in the *Evidence-Based Practice Guidelines for Clinical Diabetology in Traditional Chinese Medicine (2016)* and the Chinese Guidelines for the Prevention and Treatment of Type 2 Diabetes Mellitus (2020 edition). 2) Age: 18 to 70 years. 3) Newly diagnosed with Type 2 Diabetes Mellitus and have not received any prior treatment for the condition. 4) Willing to participate in the study, capable of providing signed informed consent, and able to undergo the treatments and all required examinations. (3) Inclusion Criteria for Healthy Volunteers: 1) Normal blood glucose levels. 2) Age: 18 to 70 years. 3) Body Mass Index (BMI) between 18.5 and 23.9 kg/m², inclusive. 4) No history of chronic diseases and no use of any medications for therapeutic purposes within the past two years.

Exclusion criteria

Exclusion criteria: 1) Patients previously diagnosed with Type 1 Diabetes Mellitus, Gestational Diabetes Mellitus, Secondary Diabetes, or other specific types of diabetes. 2) Having taken or currently taking glucose-lowering drugs or other prescription medications within the last 2 months. 3) Patients complicated with severe cardiovascular or cerebrovascular diseases, other severe comorbidities, psychiatric disorders, or behavioral inhibition. 4) Individuals who experience fear or apprehension towards acupuncture therapy and refuse to receive it. 5) Women who are pregnant, lactating, or planning to become pregnant in the near future. 6) Individuals currently participating or who have participated in other clinical trials within the past 3 months. 7) Those who have used high-dose probiotics within the past 6 months or used topical/local antibiotics or other agents that may affect oral metabolite detection within the past 7 days. 8) Individuals who are ineligible for MRI scanning for any reason. 9) Patients deemed by the investigators to be unsuitable for participation in the study.
