Inclusion criteria

Inclusion criteria: Participants must have given their written informed consent, and their ages and genders should be evenly distributed between the following decades of life: 18-29 years, 30-39 years, 40-49 years, 50-59 years, 60-69 years, and finally >70 years. Participants must be able to understand the full nature and purpose of the study, including possible risks and adverse effects.

Exclusion criteria

Exclusion criteria: Presence of any evident skin disease at the sampling site.

MeasureTime frame
To ultimately develop a robust DNA methylation–based predictor of skin cancer risk, ultra-low-input methylation profiling using Infinium EPIC arrays will be performed to generate genome-wide methylation profiles from a large cohort of healthy epidermis samples from participants. In addition, the aim is to train a specific DNA methylation clock for tape-lift samples that could detect a potential acceleration of molecular age, which may be associated with an increased risk of developing keratinocyte carcinomas.

MeasureTime frame
Image data, skin hydration measurements, and questionnaire data will be analyzed using a combination of advanced analytical methods. In addition, further molecular profiles can be generated from the collected biological material, such as transcriptomic, proteomic, and/or skin microbiome data. For the analysis of image, hydration, and questionnaire data in conjunction with the epigenetic data, both multimodal models and single-input methods will be employed. Furthermore, differential DNA methylation analyses will be conducted between the generated healthy methylomes, precancerous samples, and various types of epidermal carcinomas.
