Inclusion criteria

Inclusion criteria: - Voluntary participation confirmed with signature (see patient or volunteer information and consent) - At least 18 years old - BMI over 18.5 kg/m2 - Memory performance at least 1.3 standard deviation below the norm (CERAD) - MMSE = 26

Exclusion criteria

Exclusion criteria: - Iron deficiency - Pregnancy/breastfeeding - Hemophilia - Taking anticoagulants - Blood donation in the last 8 weeks before the study day - Current presence of serious mental illness that could explain the memory deficit (major depressive episode, schizophrenia, severe substance abuse or dependence)

cognitive performance Various cognitive tests from the Consortium to Establish a Registry for Alzheimer's Disease (CERAD; German version) were used to assess cognitive performance. The CERAD test, which comprises a total of 11 subtests, includes five tests that are relevant for this study: The Animal Fluency Test (AFT) and the Semantic Fluency Test (SFT) measure executive functions and linguistic performance. They were combined into one “fluency” category. The Immediate Recall (IR) and the Delayed Recall (DR) both measure episodic memory over different time intervals. Finally, the Trail Making Test - Part B (TMT-B) was included to measure executive functions, in particular cognitive flexibility and attention.

mitochondrial function Mitochondrial function was assessed in peripheral blood mononuclear cells (PBMCs). The ATP content of the cells was determined using a luminescence kit (6016949, Perkin Elmer, Rodgau-Jügesheim, Germany). The activity of citrate synthase (CS) was measured by the conversion of DTNB to TNB in a self-developed assay. CS and ATP were measured using a ClarioStar Plate Reader (BMG Labtech, Ortenberg, Germany). To determine the complex activity of the respiratory chain, an Oxygraph-2k respirometer (Oroboros Instruments, Innsbruck, Austria) was used.
