Inclusion criteria

Inclusion criteria: pregnancy with one fetus after the 20th pregnancy week

Exclusion criteria

Exclusion criteria: fetus has genetic defects like trisomy; multiple pregnancy

Primary

Measure	Time frame
The primary aim of this study is the immunoligical characterization of preeclampsia. For that we will measure negative checkpoint regulators (PD-L1, VISTA), marker for antigen presentation (MHC-II, CD74) and other immune marker via qRT-PCR on fixed placenta tissue, frozen placenta tissue and isolated macrophages from cord blood and maternal blood. In addition we will stain the marker via immunohistochemistry on the fixed placenta tissue. From the frozen placenta tissue, macrophages and trophoblasts can be isolated. And the stem cells from cord blood or monocytes from maternal blood can be differentiated into macrophages. These macrophages will be used for functional tests of immune regulation of preeclampsia. For that we will perform migration-/invasionassays, immunecytochemistry, flow cytometry of the mentioned marker, immune precipitation and westen blots as well as IL-2/IL-6 elisas to measure the stimulation of the macrophages. For that we would not only use cell culture, but cell co-culture of macrophages + trophoblasts/T-cells.	—

Measure	Time frame
The additional aim of this study is the isolation and characterization of exosomes from maternal blood. We will sarch for immunological marker for a better and earlier diagnosis and immunological prognostic marker of the preeclampsia.	—
