Inclusion criteria

Inclusion criteria: • Patients with a histologically approved EoE or strong suspicion of EoE, who receive an upper endoscopy; • Hemoglobulin of at least Hb = 10 g/dl • Age > 3-20 years • Approval of patient (if > 6 years) or parents

MeasureTime frame
Following the assumption that T-cells play a crucial role in the multistep pathogenesis of EoE, we would like to characterize the tissue-resident T-cells in the Esophagus. In a prospective clinical study differences in the T-cell cohort will be analyzed longitudinally at the timepoint of diagnosis, after medical therapy and elimination diet. Aims and objectives: 1) Describing T-cell subpopulations and characterizing their function for eosinophilic inflammation and resolution in EoE. 2) Prospective characterization of the local T-cell receptor repertoire in the course of the disease. 3) Correlation of the T-cell immunobiology with local environmental factors like food allergens and the mucosa associated microbiome, as well as medical therapy and clinical Response (disease activity index, histopathology).

MeasureTime frame
In the long run we aim at developing a novel therapeutic approach, which aims at T-cell immunoregulation and modulation of the gastrointestinal dysbiosis, using targeted alimentary or micbrobial interventions (e.g. nutrition, probiotics, selective antibiotics, microbial transfer). Additionally our objective is to identify novel biomarkers for diagnostic and therapeutic Response, which allow to stratify patients according to a targeted therapy.
