Conditions

Experimentally induced contact eczema

Interventions

Group 1: We will establish experimental contact eczema as chronic itch model in volunteers using diphencyprone. This model provides localized inflammatory ongoing itch that is ideally suited to study

Inclusion criteria

Inclusion criteria: Caucasian volunteers

Exclusion criteria

Exclusion criteria: Patients with chronic itch; Volunteers taking antipruritic drugs or having an infection at the potential biopsy area.

Design outcomes

Primary

MeasureTime frame
Morphological parameters: epidermal and dermal nerve fiber density, caliber deviations, branching, characterization of nerve fibers (myelin, neuropeptides, non-peptidergic markers, receptor expression). Functional parameters: pruritus intensity, spontaneous and evoked by electrical stimulation as well as diphencyprone application. Pruritus/pain intensity evoked by injection of lysophosphatidylic acid (LPA), BAM 8-22 (bovine adrenal medulla peptide 8-22), Beta-Alanin and Interleukin-31 after NGF sensitization. Immunostaining: expression of pruritic mediators. Gene expression: cutaneous global gene expression profiles, differential expression profiles. Microdialysis: lysophosphatidylic acid (LPA) analysis.
