Inclusion criteria

Inclusion criteria: - endurance-untrained (VO2max <40 ml/min *kg) - declaration of consent - Age between 18 and 29 years - medical certificate for sports aptitude

Exclusion criteria

Exclusion criteria: - injuries, which do not allow the participation in the sport program - chronic diseases, especially intestinal diseases (eg. ulcerative colitis, celiac disease) - eating disorders - regular medication intake

Primary

MeasureTime frame
The microbiota represents the primary endpoint. Microbial composition (proliferation favourable bacteria species, such as lactic bacteria), bacterial metabolites (increasing short-chain fatty acids) and intestinal barrier (strengthening of the intestinal mucosa) will be investigated as markers for a „healthy“ microbiota. These are the analysed parameters in detail: Fecal total colony count, Luminal protective flora, Immunmodulating flora, Proteolytic flora, Mukonutritiv flora, Short chain fatty acids, Yeast fungus diagnostics, Mold fungus diagnostics, Stool characteristics, Digestion residues (fat, iso-fatty acids, total fatty acids, nitrogen, water, bile acid), Pancreas specific elastase, Secretory immunoglobulin A, Eosinophile Protein X, ß-Defensin, Zonulin, Calprotectin, Alpha-1-Antitrypsin, Lactoferrin, Lysozyme. The examination will be carried out before, during and after the intervention programme: T0: befor the intervention T1: after two-week-rest-period T2: after four-week-endurance-/probiotic-period T3: after four-week-probiotic-/endurance-period Stool samples will be submitted for the investigation of the fecal parameters. The test subjects get prepared material from the "MVZ Institut für Mikroökologie GmbH". This includes a stool collector and a shipping box. The specimen collection is made at eight different points of the stool. The stool sample will be send to the "MVZ Institut für Mikoökologie GmbH" (not just before or on the weekend). If the shipment is not made immadiately after sampling, the stool sample will be stored in the refrigator (not freezer). The analysis is carried out by the MVZ Institut für Mikroökologie GmbH with "KyberKompaktPRO-Darmflora-Diagnostik", "KyberPlus-Stuhldiagnostik" and "Gaschromatographie, Enzyme-Linked-Immuno-Sorbent-Assay".

These secondary endpoints are also collected: - variations of different biomarkers in the blood ? reduction of cytokines (IL-6, IL-8, IL-1ß, TNF-a) ? regulation of serum ferritin, vitamin D - reduction of selected inflammatory parameters in the saliva (TNF-a, IL-6, IL-8, IGN-y) - lifestyle and physical activity out of the programme - inclusion of macro- and micronutrients during the endurance programme, or relationships between dietary composition and microbial modifications (effects of carbohydrates, proteins, fats, fiber, water, vitamins, trace minerals) These parameters are also collected at the four measurement times: T0: before intervention T1: after a two-week rest period T2: after a four-week endurance-/probiotic period T3: after a four-week probiotic-/endurance period The blood sampling is performed under medical supervision. One blood tube is used for a small blood count and one serum tube is used for the other blood parameters. The blood will be centrifuged for ten minutes at 4000 rotations, pipetted and frozen at -80 °C (until further examinations). The blood test is made with a hematology equipment. The serum samples are analyzed by: ELISA Kit EHFTL, Multiplex ELISA HADCYMAG-61-K, EIA-5396. The saliva is collected by Salivettes from Sarstedt. The diagnosis of the salivary parameters (TNF-a, interleukin-6, interleukin-8, interferon-gamma) is performed by multiplex ELISA Multiplex ELISA HCYTOMAG-60K. Based on a modified IPAQ (International Physical Activity Questionnaire), lifestyle and sports activities (beyond the intervention programme) are determined. Nutrition protocols are used to document food intake (quality and quantity) for 5-7 days. The protocols will be analyzed with specific software (DGE-professional).
