Inclusion criteria

Inclusion criteria: 1. Aged 18–85 years, male or female. 2. Patients diagnosed with amyotrophic lateral sclerosis (ALS) according to the Gold Coast criteria.

Exclusion criteria

Exclusion criteria: 1. Women who are pregnant, breastfeeding, or planning pregnancy. 2. Other neuromuscular diseases: patients with peripheral neuropathy, myositis, or other inherited muscular diseases, to avoid confounding spatial-temporal transcriptomic results. 3. Systemic diseases: patients with severe autoimmune diseases, active infections, or malignant tumors (which may interfere with immune profiling). 4. Drug interference: administration of antibiotics, probiotics, or prebiotics within 4 weeks prior to screening (which severely affects metagenomic data). 5. Severe gastrointestinal diseases. 6. Participation in other clinical trials of investigational drugs, or use of special medications that may affect metabolomic profiles. 7. Skin infection at the puncture site or severe spinal deformity that precludes lumbar puncture. 8. Refusal to participate in this study. 9. Any other conditions judged by the investigators to render the patient ineligible for study participation.
