Inclusion criteria
Inclusion criteria: 1. Inclusion criteria of KOA patients: (1) Diagnosis of KOA according to the American College of Rheumatology (ACR) criteria; (2) Radiographic severity graded as Kellgren-Lawrence (KL) grade >= II; (3) Pain intensity >= 3 on a 0-10 Numeric Rating Scale (NRS); (4) Duration of knee pain >= 3 months; (5) Aged between 45 and 70 years; (6) Right-handed; (7) No history of cognitive impairment, psychiatric disorders, or neurological diseases; 2. Inclusion criteria of HCs: (1) No history of knee pain, arthritis, or other chronic pain disorders; (2) Matched to KOA patients by age and sex; (3) Right-handed; (4) Knee joint X-ray films show a Kellgren-Lawrence (KL) grade <= ? (or no definite radiological evidence of osteoarthritis); (5) No history of cognitive impairment, mental illness, or central nervous system diseases, and ability to cooperate with resting-state functional magnetic resonance imaging (rs-fMRI) scanning and various assessments; (6) No use of non-steroidal anti-inflammatory drugs (NSAIDs), opioids, or other drugs affecting central pain processing within 1 month prior to enrollment; (7) No contraindications to magnetic resonance imaging (MRI) examination (e.g., metallic implants, claustrophobia, etc.); (8) Ability to understand and sign the written informed consent form.
Exclusion criteria
Exclusion criteria: 1. Exclusion criteria of KOA patients: (1) History of knee surgery within the previous 6 months; (2) Presence of other inflammatory arthritis, fibromyalgia, or other chronic pain conditions; (3) Major psychiatric disorders or neurological diseases; (4) Severe systemic comorbidities;(5) Use of centrally acting analgesics, opioids, benzodiazepines, or antidepressants within 2 weeks before scanning; (6) Excessive head motion during fMRI acquisition (> 2 mm translation or > 2° rotation); (7) Structural brain abnormalities. (8) MRI contraindications. 2. Exclusion criteria of HCs: (1) History of systemic inflammatory diseases, traumatic knee injury, or chronic pain; (2) Major physical, psychiatric, neurological, or cognitive disorders; (3) Use of centrally acting medications within 2 weeks before MRI scanning; (4) MRI contraindications or structural brain abnormalities detected on MRI; (5) Head motion > 2 mm translation or > 2° rotation during MRI scanning.
