Design outcomes

Primary

MeasureTime frame
Clinical data analysis (age, gender, duration of diabetes, HbA1c, eGFR, urine protein levels, serum creatinine, BUN, etc.);Histopathological analysis (using HE staining, PAS and Masson staining to observe the degree of glomerulosclerosis, mesangial matrix expansion, and tubular interstitial fibrosis);Protein localization and expression analysis (using immunohistochemistry (IHC) to detect the expression intensity and localization of proteins such as MECP2, SPI1, NLRP3, LC3, PINK1, and P62, with scoring based on staining intensity and positive area);Observation of mitochondrial morphology and structure (using transmission electron microscopy to observe the quantity, structure, cristae morphology, presence of vacuolization, and other pathological changes of mitochondria, and to assess autophagosome formation);
