Inclusion criteria
Inclusion criteria: 1. Age range: 18–60 years. (healthy individuals); 2. Health status: No acute infections (normal body temperature, CRP, and PCT levels). No chronic inflammatory diseases (e.g., rheumatoid arthritis, systemic lupus erythematosus). No history of immunosuppressant use (e.g., glucocorticoids, biologics). (healthy individuals); 3. Laboratory parameters: Complete blood count: White blood cell count: (4–10) ×10^9/L Monocyte proportion: 2–8% (within reference range). Infection biomarkers: PCT < 0.05 ng/mL CRP < 5 mg/L. (healthy individuals); 4. Lifestyle requirements: No vaccination or significant immune-stimulating events (e.g., surgery, trauma) within 1 week prior to blood sampling. (healthy individuals); 5. Diagnostic Criteria: Meeting the Sepsis-3.0 criteria, defined as infection + SOFA score =2 points. (for septic patients); 6. Disease Course Requirements: Early sepsis (samples collected 24–72 hours post-diagnosis to avoid interference from the immune paralysis phase). No advanced life support (e.g., ECMO) administered to preserve cell viability. (for septic patients); 7. Age: 18–70 years. (for septic patients);
Exclusion criteria
Exclusion criteria: 1. Active Infections: Respiratory tract/urinary tract infections, or viral carrier states (HBV, HCV, HIV, or syphilis seropositivity). (healthy individuals); 2. Immune-Related Disorders: Autoimmune diseases, active allergic diseases, or history of malignancy. (healthy individuals); 3. Special Physiological Status: Pregnant or lactating women (due to hormonal influences on immune cell phenotypes). (healthy individuals); 4. Medication Interference: Recent use (within 3 months) of immunomodulatory agents or long-term antibiotics. (healthy individuals); 5. Underlying Immune Disorders: Autoimmune diseases (e.g., SLE, RA), hematologic malignancies, or HIV infection. (for septic patients); 6. Immunosuppressive Therapy: Current or recent (within 1 month) use of chemotherapy, high-dose corticosteroids (>20 mg/day prednisone equivalent), or other immunosuppressants. (for septic patients); 7. Complex Comorbidities: • Cirrhosis (Child-Pugh class C) or end-stage renal disease (eGFR <15 mL/min/1.73 m^2); • Post-solid organ transplantation. (for septic patients); 8. Sample Interference Factors: • Leukocyte count <1.0×10^9/L (severe bone marrow suppression); • Hemolysis or coagulation disorders resulting in unqualified samples. (for septic patients);
