Major Depressive Disorder

Interventions

rTMS group:Accept 10 sessions of 10Hz rTMS stimulating the right precuneus (1/day, 5days/week, 2 weeks), each session with 3000 pulses (4s stimulation, 16s interval, 25 min)

Sham group:Accept 10 sessions of sham stimulations with 10Hz on the right precuneus (1/day, 5days/week, 2 weeks), each session with 3000 pulses (4s stimulation, 16s interval, 25 min)

Inclusion criteria

Inclusion criteria: 18–65 years old; Han nationality; Patients with a DSM-5 clinical criteria for MDD and a HAMD-17 score =18; Right-handed; Years of education = 9 years; Uncorrected or corrected visual acuity is normal in both eyes

Exclusion criteria

Exclusion criteria: Demonstrated psychotic symptoms; Suicidal risk (as defined by a score =3 on HDRS item 3); Exhibited a severe and unstable physical illness; A history of head injury, stroke, brain tumor, seizure disorder or family history of seizure disorder and so on; Diagnosed with other mental disorders; Pregnancy or lactation; Prior treatment with TMS, tACS, tDCS, MECT in the past 6 months; Personality disorders such as antisocial personality and borderline personality; Taking more than 2 mg lorazepam (or an equivalent) or any anticonvulsant; Equal or more than two adequate anti-depressant trials had failed, or taking two or more than two kinds of antidepressants; Patients with contraindications to rTMS intervention or MRI scanning: a. cardiac pacemakers, artificial metal valves, etc.; b. Intracranial postoperative arterial enclanglement; c. There are ferromagnetic substances such as shrapnel and metal foreign bodies in the body; d. Metal prostheses, metal joint replacements, metal braces, etc.; e. Insulin pumps, nerve stimulators, etc.; f. claustrophobia; g. Tattoos or eyebrow tattoos (containing metallic substances) in the last two weeks.
