Inclusion criteria

Inclusion criteria: 1. the patients receiving mechanical ventilation requires sedation in ICU; 2. The expected duration of mechanical ventilation longer than 24H; 3. aged 18 to 80 years; 4. agreed to participate in the study, and informed consent.

Exclusion criteria

Exclusion criteria: 1. allergic to opioid 2. patients with shock: MBP 3 months); 7. alcohol or drug dependent patients; 8. patients with neuromuscular diseases; 9. disturbance of consciousness (trau matic brain injury, stroke, intracranial infection, dementia, etc.); 10. liver dysfunction: ALT or AST> normal 5 times; 11. body mass index BMI (weight kg/height m2) is not 18 to 30; 12. patients with organic heart disease preopertation (such as sick sinus syndrome, atrioventricular block, etc.); 13. severe dehydration, electrolyte imbalance; 14. with severe renal dysfunction and severe disturbance water and salt metabolism.
