Brief summary
This study aims to investigate the effects of fat content in a mixed-nutrient meal on muscle and adipose tissue microvascular blood flow (MBF) in healthy humans. We will compare the effects of low-fat vs high-fat content in a mixed-nutrient meal ( a liquid milkshake) on post-meal skeletal muscle and adipose tissue MBF healthy humans in a randomised crossover design.We will also measure gut-derived hormones and perform a correlation analysis with the MBF response. Hypothesis: We hypothesise that the low-fat mixed-nutrient meal will increase muscle and adipose MBF after the meal in healthy individuals, while the high-fat meal mixed-nutrient will not show this increase in MBF responses. We also hypothesise that levels of gut-derived hormones in the plasma will be correlated to MBF responses. Expected outcomes and significance: We aim to show that fat content within a mixed-nutrient meal is an important factor determining tissue MBF response and can affect the risk of cardiometabolic disease.
Interventions
This study aims to investigate the effects of fat content in a mixed-nutrient meal on muscle and adipose tissue microvascular blood flow (MBF) in healthy humans. We will be using a pre-tested low-fat meal (control meal) that has been demonstrated to increase microvascular responses in healthy people. This meal provides 1306 kJ of energy and contains 38 g of carbohydrates, 21 g of protein and 5 g of fat.. We will compare its effects to that of a high-fat meal that provides 1680 kJ of energy and contains 38 g of carbohydrates, 21 g of protein and 15 g of fat. This study will be a repeated measures randomized cross-over design with participants coming into the laboratory for 2 visits (7 to 10 days apart). During each visit participants will consume one of the two meals (in a random order) and will undergo post-meal blood flow and metabolism assessment. The participants will be blinded to the meal type. The participants will consume the meal infront of the researchers.

Inclusion criteria
Age 18-45 years. Body Mass Index (BMI) 18-24.9 Kg/m2. Normotensive (seated brachial blood pressure <140/90 mmHg). Have no dietary allergies, food intolerances, or dietary restrictions. Have given signed informed consent to participate in the study.
Exclusion criteria
People do not meet the inclusion criteria outlined above. People tell us they cannot read or understand the English language. Smoking status – People who are regular smokers in the last 10 years and/or >10 years of smoking history, or who use vapes. People have a history of severe liver disease, any vascular or cardiovascular disease, or diabetes. Pregnant/lactating women. People with a family history of type 2 diabetes (i.e., a parent with type 2 diabetes). People already on a high-fat diet (e.g., keto diet), or a vegan/vegetarian diet. People who have known food allergies, intolerances, or dietary restrictions. People who suffer from any respiratory issues (such as asthma). People who have previously had a reaction to the Definity contrast agent. People who exercise more than the current guidelines for physical activity (>5 hours of moderate intensity physical activity or > 2.5 hours of vigorous intensity physical activity per week).
