Brief summary

This study will test whether drinking beetroot or blackcurrant beverages with a carbohydrate-containing meal can reduce the rise in blood glucose and insulin after eating. Healthy adults will attend several study visits where they consume toast with either a beetroot, blackcurrant, or placebo drink, followed by blood sampling over 2.5 hours. The purpose is to understand whether naturally occurring compounds in these foods, such as nitrate and polyphenols, may help improve short-term metabolic responses after meals. We hypothesise that beetroot and blackcurrant beverages will reduce post-meal glucose and/or insulin responses compared with placebo.

Interventions

18 healthy participants will complete glucose tolerance tests, consuming 75g of carbohydrate (toast) with either a beetroot, blackcurrant or placebo beverage (total 75g carbohydrate). The beetroot beverage will contain 600mg of nitrate, and the blackcurrant beverage 300mg of polyphenols. Both beverages are made to match sugar content (42.9 grams), as well as placebo. The intervention will be administered by the randomisation officer (either a Nutrition PhD student or Lab Manager) and participant

18 healthy participants will complete glucose tolerance tests, consuming 75g of carbohydrate (toast) with either a beetroot, blackcurrant or placebo beverage (total 75g carbohydrate). The beetroot beverage will contain 600mg of nitrate, and the blackcurrant beverage 300mg of polyphenols. Both beverages are made to match sugar content (42.9 grams), as well as placebo. The intervention will be administered by the randomisation officer (either a Nutrition PhD student or Lab Manager) and participants will be asked to consume the meal within 10minutes. The participants will be supervised by the randomisation whilst eating to ensure all food and beverage is consumed. Blood samples will be taken by cannulation and finger prick sampling at baseline and after 5, 15, 30, 45, 60, 90, 120 and 150 min following ingestion. Each testing session will last 3hrs total. Each testing session will occur at least 48hr apart to allow for washout of bioactive ingredients. The efficacy of each beverage will be assessed by comparing incremental area under the curve (iAUC) for glucose and insulin sensitivity via HOMA IR index against placebo.
